Offer Description Osteoarthritis is the most common debilitating joint disease, resulting from inflammation of the synovial membrane and destruction of the cartilage. Osteoarthritis is a debilitating and painful disease whose causes are poorly understood, and current treatments are mainly symptomatic. To better understand the development of the disease and test new therapeutic strategies, there is an urgent need to develop new in vitro models of osteoarthritis. 3D bioprinting is a promising technology for producing osteoarthritic tissue in vitro in a reproducible and automated manner. However, most printable biomaterials lack tunability in terms of composition and mechanical properties, and current bioprinting processes do not allow for high shape fidelity. In this context, we hypothesise that the development of new bioprinting tools will enable the production of biologically relevant joint tissues for modelling osteoarthritis in vitro. In particular, we will develop ultra-tunable hydrogels enabling the printing of high-resolution objects. We will use these printable hydrogels to design cartilage tissues in vitro. The biological relevance of the engineered tissue will be validated, with a view to the longer-term development of a complete in vitro osteoarthritis joint model.
Responsibilities Develop a series of ultra-tunable biomaterials: Optimize the physicochemical properties of hydrogels for printability
Validate the cytocompatibility of the printable hydrogels
Design a bioprinted cartilage tissue: Develop a bioprinted model of cartilage
Characterize the model both mechanically and biologically
